چکیده

Planar cell polarity (PCP) refers to the collective polarization of cells along a tissue plane, such as ordered alignment body hairs in skin. Disruption PCP leads developmental disorders heart, neural tube, inner ear and A hallmark feature is asymmetric localization core proteins Fz6 Vangl2 at an intercellular junctional complex organized by cadherin family member Celsr1. Our studies Celsr1 mouse revealed that functions not only trans-adhesive molecular bridge, but also organizer asymmetry junctions through lateral, cis-interactions dimerization. These reveal key roles for adhesion vertebrate development. Interestingly, mutations occur patients with epidermal-derived tumors carcinomas malignant melanomas, defects spina bifida congenital heart defects. We hypothesized these perturb establishment function impairing adhesion. To test this possibility, WT various mutants was assessed using combination assays, advanced optical imaging, vivo murine genetic approaches. identified repeats were defective determined ability accumulate cell-cell borders form stable adhesive interactions fluorescence recovery after photobleaching. melanoma result variants display significantly reduced function. In addition loss-of-function mutants, we human gain-of-function activity. Overall, our findings indicate organize coordinate suggest signaling are altered both skin cancers.

Planar Cell Polarity Protein Celsr1 Regulates Endothelial Adherens Junctions and Directed Cell Rearrangements during Valve Morphogenesis

Planar cell polarity (PCP) signaling controls tissue morphogenesis by coordinating collective cell behaviors. We show a critical role for the core PCP proteins Celsr1 and Vangl2 in the complex morphogenetic process of intraluminal valve formation in lymphatic vessels. We found that valve-forming endothelial cells undergo elongation, reorientation, and collective migration into the vessel lumen ...

Aberrant regulation of planar cell polarity in polycystic kidney disease.

Mutations in PKD1, which encodes polycystin-1 (PC1), contribute to >85% of cases of autosomal dominant polycystic kidney disease (ADPKD). The planar cell polarity (PCP) pathway is necessary for the oriented cell division and convergent extension that establishes and maintains the structure of kidney tubules, but the role of this pathway in the pathophysiology of ADPKD is incompletely understood...

Plasticity of both planar cell polarity and cell identity during the development of Drosophila

Drosophila has helped us understand the genetic mechanisms of pattern formation. Particularly useful have been those organs in which different cell identities and polarities are displayed cell by cell in the cuticle and epidermis (Lawrence, 1992; Bejsovec and Wieschaus, 1993; Freeman, 1997). Here we use the pattern of larval denticles and muscle attachments and ask how this pattern is maintaine...

Cell polarity triggered by cell-cell adhesion via E-cadherin.

Cell polarity is orchestrated by numerous extracellular cues, and guides events such as chemotaxis, mitosis and wound healing. In scrape-wound assays of cell monolayers, wound-edge cells orient their centrosomes towards the wound, a process that appears to depend on the formation of new cell-extracellular-matrix adhesions as cells spread into the wound. In direct contrast to scrape-wounded cell...

Regulation of Planar Cell Polarity by Smurf Ubiquitin Ligases

Planar cell polarity (PCP) is critical for morphogenesis in metazoans. PCP in vertebrates regulates stereocilia alignment in neurosensory cells of the cochlea and closure of the neural tube through convergence and extension movements (CE). Noncanonical Wnt morphogens regulate PCP and CE in vertebrates, but the molecular mechanisms remain unclear. Smurfs are ubiquitin ligases that regulate signa...
